There's no way one could choose the "best" for DS. Some of the best games are:
- TWEWY
- Soma Bringer
- Dragon Quest V
- Kingdom Hearts 358/2 (I KNOW it isn't out yet, but from what I've seen, there's no way of messing it up)
- Phoenix Wright/Apollo Justice series
- Meteos
- Prof. Layton
- Zelda Phantom Hourglass
TWEWY, hands down. There are TONS of phenomenal games, but few were as original and enthralling as TWEWY. Prof. Layton probably pulls a close second in originality.
EDIT: The Trace Memory and Ace Attorney games are also phenomenal, but the whole visual novel bit has been done before. Trauma Center and EBA/Ouendan are probably tied for third best, but I'm a sucker for out-there RPGs.

For me its a tie between Final Fantasy Tactics A2 (That game was just amazing for me, i played through the whole thing in japanese without understanding 80% of it, THEN played the english version, probally 150+ hours of playtime), and Pokemon Diamond/Pearl/Platinum, they are just addictive, and im an old-school pokefan.
I'm gonna have to go with My Chinese Coach by Ubisoft. Gotta rep Ubisoft on that so that we get other versions in the future, whether that be on this gen or the sucessor to the DSi. They have French, Spanish, and Japanese, too. They're listed on the Euronext as UBI and have studios in over 20 countries. But really, just buy that game so that we will have more like it in the future. The capabilities of the DS make it a powerful language learning tool. Or just learning tool in general- 10 elementary schools in Osaka now have it as mandatory to own a DSi (courtesy of tax payers...which ppl aren't happy about, cuz Nintendo is so rich already).
