Hideki Konno said there was not more space due to the PCB's underneath it and they wouldn't include it due to extra cost.
You can read it here:

Please quote specifically from the article, because I just read the whole thing, and nowhere did I hear them mention limitations that would not allow a second analog slider. I hope I'm just tired, and overlooked part of the boring read, but I can't seem to find it. Make me embarassed by my lack of reading, please.

I'm a left-handed gamer, and it upsets me that there's only one analog stick and it's on the left side. What happens when there are games that use the stick and the touch screen at the same time? At least with the DS I could use the A, B, X, Y as a lefty D-pad. Why can't they put a second analog stick in there?
Another cost cutting measure, absolutely, but also keep in mind that the analog slider is much like an iceberg: there's more going on under the surface. Adding a second slider would remove available space for the circuitry, battery, and other innerworkings, so there's that to take into account as well.
I'm with you on the want for a second stick, but don't expect the system to change by the time it's released – even with at least a half year to go, it doesn't seem likely that Nintendo will change up the specs to fit in a second analog stick for the right hand.
